Anti-Inflammatory Effects of Schisandrin A on Lipopolysaccharide-Induced RAW264.7 Cells in Mice
Objective: To probe into the anti-inflammatory effects of schisandrin A on lipopolysaccharide-induced RAW264.7 cells in mice. Methods: Effects of schisandrin A on proliferation
apoptosis and phagocytosis of lipopolysaccharide-induced RAW264.7 cells in mice and levels of interleukin (IL)-1β
tumor necrosis factor-α (TNF)-α
prostaglandin E2 (PGE2) and cyclooxygenase (COX)-2 levels were detected. The proliferation of RAW264.7 cells was measured by CCK8 method and the phagocytosis of RAW264.7 cells was detected by neutral red reagent. IL-1β
TNF-α
PGE2 and COX-2 were determined by enzyme-linked immunosorbent assay (ELISA). Annexin V-FITC/PI double staining was used to detect the effects of schisandrin A on apoptosis of RAW264.7 cells after induction. Results: Schisandrin A could significantly inhibit the proliferation
phagocytosis and apoptosis of lipopolysaccharide-induced RAW264.7 cells in mice. ELISA results showed that schisandrin A could inhibit the secretion of IL-lβ
TNF-α
PGE2 and COX-2 in RAW264.7 cells. Conclusion: Schisandrin A has significant anti-inflammatory effects on lipopolysaccharide-induced RAW264.7 cells in mice
and its mechanism may be related to inhibiting the proliferation and phagocytosis of inflammatory cells
reducing the release of inflammatory factors
and reliefing the lipopolysaccharide-induced apoptosis.
